Randomized Cross over Trial of PBMCs Genomics In Vivo Using Different Dialyzer Membranes.

We studied genome wide gene expression profiles of PBMCs of patients undergoing hemodialysis comparing semi-synthetic (SEMI-SYN) versus full- synthetic (FULL-SYN) dialyzer membranes. Special focus was put on genes involved in inflammatory processes, cell differentiation, as well as cell adhesion.

Explorative and statistical analysis was performed with the TIGR Multi Experiment Viewer (MeV) and the Bioconductor module of the R software package. Differentially expressed genes were further analyzed on a functional level using the PANTHER classification system. Pathways holding a significant number of genes were additionally determined.

A total of 265 differentially regulated genes were identified. 152 genes showed increased expression in the SEMI-SYN group and could be classified to the biological process categories immunity and defense, blood clotting, and cell surface receptor mediated signal transduction. Key members of the integrin and the TGF-beta signaling pathways were also among the SEMI-SYN upregulated genes. The 113 genes upregulated in the FULL-SYN group on the other hand were mainly involved in protein biosynthesis and nucleoside metabolism.

These data provide evidence for activation of inflammation in the SEMI-SYN but not FULL-SYN membranes.
